By default, WordPress will use the firstname.lastname@example.org e-mail address and “WordPress” as e-mail sender information for e-mails such as “user created”, “password reminders” and more. This is fairly anonymous and is probably not the sender for your e-mails that you want. It might even be the cause for e-mails “getting lost” or ending up in the spam folder.
The HW Override Default Sender plugin allows you to change the default WordPress e-mail sender information (including Reply-Path). It works for standard WordPress installations as well as multi-site installations. After installing and activating this plugin it will use the information you already provided in your WordPress settings unless something else has been specified (see FAQ below).

No. It will only intervene if it detects that the e-mail about to be sent has the WordPress default sender information. It means that existing plugins, filters or in other way specified e-mail sender information will be untouched and still work as expected.
It will always override the Reply-Path header if not prohibited by your server configuration. It will do so by setting the Reply-Path e-mail address to the same address as the From value.
